Purpose
- To identify sickle cell disease and sickle cell trait.
Patient preparation
Explain to the patient that this test helps detect sickle cell disease. Inform him
that he needn't restrict food or fluids, pell him the test requires a blood Sample; who will perform the venipuncture and when; and that he may experience transient discomfort from the needle puncture and the pressure of the tourniquet. Reassure him that collecting the sample takes less than 3 minutes. Check patient history for blood transfusion within the past 3 months.
Procedure
Perform a venipuncture, and collect the sample in a 7 ml lavender-top tube.
Precautions
Completely fill the collection tube, and invert it gently several times to adequately mix the sample and the anticoagulant. Don't shake the tube vigorously, since hemolysis may result,
Values
Results of this test are reported as positive or negative. A normal, or negative, test suggests the absence of Hgb S.
Post-test care
If a hematoma develops at the venipuncture site, apply warm soaks.
Interfering factors
- Hgb concentration under 10%, elevated Hgb F levels in infants under age 6 months, and blood transfusion within the past 3 months may produce false-negative test results.
- Failure to use the proper anticoagulant in the collection tube, to completely fill the tube, or to adequately mix the sample and the anticoagulant may interfere with accurate determination of test results.
- Hemolysis due to rough handling of the sample may affect test results.
