Major Uses: Chloroquine is used for the treatment and prevention of malaria, a parasitic infection transmitted to humans by some types of mosquitoes. It can also be used in rheumatoid arthritis and some other inflammatory conditions.
How it Works: The action of chloroquine in fighting malaria is not known. It is likely to have a number of effects on various stages of the malaria parasite.
Usual Dosage: 300mg once per week for prevention of malaria in adults. Lower doses for children. (This should be taken for two weeks before, during and for four weeks after leaving a malaria area.) Treatment of established malaria requires higher doses.
Common Side-Effects: Nausea, headache and abdominal discomfort are the most common side-effects when the tablet is taken in low doses or for short periods of time. If the drug is used in the long-term a number of eye problems can occur. Opacity of the cornea can occur, but resolves if the medication is stopped. Degeneration of the retina is less common, but is not reversible and can cause permanent deterioration in vision.
Actions with other Drugs: Antacids may reduce the absorption of the drug.
Pregnancy, Breastfeeding: This medication is not safe to use during pregnancy or while breastfeeding. Definite abnormalities related to the use of chloroquine have been seen in the fetus.
Special Features, Comments: Intentional or accidental over-dosing with chloroquine can be extremely dangerous or even fatal, as in high concentrations the conduction of impulses within the heart is affected. Immediate medical attention is vital if this is suspected. Alcohol may interact with chloroquine and cause liver problems.
